ASK® URITEST 10A

PURPOSE  

     The ASK Uritest® 10A is, a urine strip contains 10 reagents, used for the detection of Leukocytes, Nitrite, Urobilinogen, Protein, pH, Occult blood, Specific gravity, Ketone Bilirubin and Glucose. The results may provide information regarding the status of carbohydrate metabolism, kidney function, liver function, acid-base balance and bacteriurea.

PRINCIPLE

   a. Leukocytes: The test is based on the detection of “esterase in leukocyte
                catalyze the hydrolysis of ester derivate”.
   b. Nitrite: The test is based on the detection of “nitrite + p-arsanilic acid ------- >
            diazonium compound”.
   c. Urobilinogen: The test is based on “Ehrlich reaction”.
   d. Protein: The test is based on the detection of “protein + tetrabromphenol blue
             -------- > yellow green and green to green blue”.
   e. pH: The test is based on double indictor (methyl red and bromthymol blue)
         principle.
   f. Occult blood: The test is based on the detection of “peroxidase-like activity of
                hemoglobin”.
   g. Specific gravity: The test is based on the detection of “the pKa change of
                   polyelectrolytes in relation to ion concentration”.
   h. Ketone: The test is based on the detection of “the reaction between acetoacetic
            acid with nitroprusside”.
   i. Bilirubin: The test is based on the detection of “bilirubin + diazotized
             dichloroaniline --------- > color complex”.
   j. Glucose: The test is based on a double sequential enzyme reaction.
            1st : glucose oxidase catalyzes the formation of gluconic acid and
                H2O2 from the oxidation of glucose.
            2nd : peroxidase catalyzes the reaction of H2O2 with a KI chromogen to
                oxidize the chromogen to colors ranging from green to brown.
